Getting rid of nucleases from purified plasmids?
The plasmid DNA may be contaminated with various cellular nucleases that can degrade the DNA when the magnesium containing restriciton digestion buffer is added. What can you do to minimize degradation of your plasmid DNA during the restriction digestion step?
